Procedure
A solution of tert-butyl 4-{1-[(5-methyl-1,3-thiazol-2-yl)methyl]-2,4-dioxo-6-phenyl-1,4-dihydrothieno[3,2-d]pyrimidin-3(2H)-yl}piperidine-1-carboxylate (980 mg; compound B12) in DCM (5 ml) and 1,4-dioxane (20 ml) is reacted with a solution of hydrogen chloride in 1,4-dioxane (2.3 ml, 4.0 M) under stirring at RT. After 1 h and after 2.5 h additional amounts of a solution of hydrogen chloride in 1,4-dioxane (4.6 ml and 2.3 ml) are added and the reaction mixture is stirred for another 10 min. All volatiles are removed under reduced pressure and the resulting residue is treated with diethyl ether. Removal of the solvent under reduced pressure gives the title compound as a solid.
