To a solution of tert-butyl 4-{1-[(2-ethyl-2H-tetrazol-5-yl)methyl]-6-(5-fluoro-2-methoxyphenyl)-2,4-dioxo-1,4-dihydrothieno[3,2-d]pyrimidin-3(2H)-yl}piperidine-1-carboxylate (435 mg; compound B57) in 1,4 dioxane (10 ml) is added a solution of hydrogen chloride in 1,4-dioxane (1.858 ml, 4.0 M). The reaction mixture is stirred for 3 d at RT. To complete the reaction an additional amount of a solution of hydrogen chloride in 1,4-dioxane (0.9 ml, 4.0 M) is added and the mixture is stirred for 1 h at 60° C. Subsequently all volatiles are evaporated and the residue is treated with DCM and after removal of the solvent under reduced pressure the title compound is obtained as a solid.
