PROCEDURE

实验过程

To a solution of 4-hydroxy-2-(1H-pyrazol-1-yl)pyrimidine-5-carboxylic acid, 8-b, (206 mg, 1 mmol) in MeCN (20 ml) were added HATU (395 mg, 1.04 mmol), Et3N (105 mg, 1.04 mmol) and ethyl methyl(4-(amino(phenyl)methyl)phenyl)phosphinate, 2-d, (289 mg, 1 mmol). The resulting mixture was stirred at room temperature for 2 hours. Then it was filtered before diluted with saturated NH4Cl solution. The mixture was extracted with ethyl acetate. The organic layer was dried over sodium sulfate and concentrated under vacuum. The residue was purified on silica gel chromatography to provide the product, 8-3, (150 mg, 31%). 1H NMR (CD3OD, 300 MHz,): δ 8.65 (s, 1H), 8.57 (m, 1H), 7.91 (s, 1H), 7.76 (m, 2H), 7.52 (m, 2H), 7.34 (m, 5H), 6.64 (s, 1H), 6.36 (s, 1H), 3.85 (m, 2H), 1.67 (d, 3H), 1.25 (m, 3H).
