1-tert-Butyl 4-methyl 3-((1-cyclopropyl-3-hydroxypropylamino)methyl)-1H-pyrrolo[2,3-b]pyridine-1,4-dicarboxylate (50 mg, 0.12 mmol) was stirred in MeOH (3 mL) with 1N NaOH (1 mL) at rt for 16 h. The reaction was neutralized with 1N HCl and concentrated in vacuo. The residue was dissolved in DMF (3 mL). HATU (47 mg, 0.12 mmol) was added, and the reaction stirred at room temperature for 2 h. The solution was concentrated in vacuo and purified by preparative HPLC eluting with a gradient of 15% to 70% Solvent B in Solvent A to give 16 mg of the title compound as an off-white solid. 1H NMR (400 MHz, CD3OD) δ 8.34 (d, 1H, J=4.8 Hz), 7.35 (d, 1H, J=4.8 Hz), 7.28 (s, 1H), 5.10 (AB q, 2H, J=76.4, 16.8 Hz), 4.08-4.20 (m, 1H), 3.59 (t, 2H, J=6.4 Hz), 2.02-2.14 (m, 2H), 1.17-1.30 (m, 1H), 0.73-0.79 (m, 1H), 0.42-0.54 (m, 2H), 0.24-0.33 (m, 1H). [M+H] found 272.
