1-(2-Chloropyridin-4-yl)-1-(2-(difluoromethyl)-6-methylpyridin-4-yl)-4-fluoro-1H-isoindol-3-amine (400 mg, 0.99 mmol), pyrimidin-5-ylboronic acid (135 mg, 1.09 mmol) and (1,1′-bis(diphenylphosphino)ferrocene)-dichloropalladium(II) (82 mg, 0.10 mmol) were mixed in THF (3 mL). Aqueous sodium carbonate (2M) (1.490 mL, 2.98 mmol) was added and the mixture was heated in a microwave reactor for 40 min at 140° C. The mixture was filtered through a pad of Celite®, eluated with DCM (15 mL) and concentrated in vacuo. Purification by preparative HPLC afforded 60 mg (14%) of the title compound. 1H NMR (400 MHz, CDCl3) δ ppm 9.26 (s, 2 H) 9.25 (s, 1 H) 8.69 (d, 1 H) 7.68 (s, 1 H) 7.50-7.58 (m, 1 H) 7.39 (d, 1 H) 7.35 (s, 1 H) 7.25 (dd, 1 H) 7.22 (s, 1 H) 7.19 (d, 1 H) 6.75-6.42 (t, 1 H) 2.56 (s, 3 H); MS (ES+) m/z 447 [M+1]+.
