Reaction Steps
Sodium hydride (14 mg, 293 umol) was added to an ice cold solution of [2-(4-chloro-phenyl)-2H-indazol-3-yl]-cyclohexyl-methanol (50 mg, 147 umol; example 10.1) in DMF (4.5 ml) under an argon atmosphere. After 30 min. methyl-4-(chloromethyl)benzoate (41 mg, 220 umol; CAS Reg. No. 34040-64-7) was added. The mixture was stirred at ambient temperature for 5 h, poured onto ice water/brine 1/1 and extracted two times with iPrOAc. The combined extracts were washed with ice water/brine 1/1 and dried over Na2SO4. After filtration the solvent was removed under reduced pressure to give the crude product as brown oil which was purified by preparative HPLC on reversed phase eluting with a gradient of acetonitrile/water to give the title compound (16 mg, 33 μmol; 22%) as yellow solid. MS: m/e=489.4 [M+H+].
