Procedure
To one liter of an aqueous solution containing glucose, MgSO4 ·7H2O and phosphate buffer at a concentration of 0.1 M, 0.005 M and 0.05 M, respectively, is added a given amount of an insolubilized glucose isomerase. The mixture thus obtained is slowly stirred at a temperature of 70° C for 60 minutes to effect the isomerization. Then, the insolubilized isomerase is separated from the reacted solution and the amount of fructose so produced is determined according to the cystein carbazol method. The activity is calculated as in 1 above.
