Procedure
10% sodium hydroxide aqueous solution (3.61 g), ethanol (5 ml), and water (3 ml) are added to 1-benzyl-6-ethoxycarbonyl-2-n-propylbenzimidazole (0.97 g) (example 51), and the solution is refluxed by heating for one hour. The acidity of the reaction solution is adjusted to pH 6 with 10% hydrochloric acid. The solution is concentrated under reduced pressure, ethanol is added to the obtained residue, and the inorganic salt is separated through filtration. The filtrate is concentrated under reduced pressure, and 1-benzyl-6-carboxy-2-n-propylbenzimidazole (0.85 g) (example 80) is obtained. 1H-NMR (DMSO-d6, δ): 0.94 (3H, t, J=7.4 Hz), 1.73-1.81 (2H, m), 2.85 (2H, t, J=7.3 Hz), 5.59 (2H, s), 7.07 (2H, dd, J=1.1 and 8.3 Hz), 7.27 (1H, t, J=7.3 Hz), 7.33 (2H, t, J=7.4 Hz), 7.65 (1H, d, J=8.4 Hz), 7.79 (1H, dd, J=1.5 and 8.4 Hz), 8.04 (1H, s).
