Reaction Steps
An aqueous solution medium containing 15% of glucose, 0.1% of potassium primary phosphate (KH2PO4), 0.04% of magnesium sulfate heptahydrate, 2% of ammonium sulfate, 100 μg/l of biotin, 200 μg/l of vitamin B1 hydrochloride, 2 ppm each of iron ions and manganese ions and 1% of "AJI-EKI" pH 7.0, was separately charged in a small glass jar fermenters in an amount of 300 ml each. After sterilization, Lys-producing Brevibacterium flavum (ATCC 21127), which had been previously grown in a bouillon slant at 30° C. for 24 hours, was inoculated on the medium. Then incubation was initiated at 31° C. About 10 hours after, penicillin was supplemented in a concentration of 6 U/ml and the incubation was continued for 18 hours. At the end of the fermentation, 3.75 g/dl of L-Lysine and 2.05 g/dl of L-glutamic acid were produced and accumulated in the medium.
