Procedure
A solution of 4M hydrogen chloride in 1,4-dioxane (3.8 mL, 15.20 mmol) was added to a solution of tofacitinib free base (4.18 g, 13.38 mmol) in acetone (40 mL). The resulting suspension was stirred at 23 to 25° C. for 6 hours, filtered and washed with acetone (2×5 mL) to yield, after air-drying, tofacitinib hydrochloride as a white solid (5.15 g). 1H NMR analysis showed a molar ratio of tofacitinib hydrochloride to acetone of approximately 1:0.88 (12.8 wt %). Water content (KF) was 0.69%.
