In 250 ml of methylene chloride was dissolved 70 g (304 mmol) of 2-(benzyloxymethyl)-4-isopropyl-1H-imidazole (3c). Then, an aqueous solution prepared by dissolving 13 g of sodium hydroxide in 160 ml of water was added at 0° C. and the mixture was stirred for 5 minutes. Then, a solution of iodine (49 g, 386 mmol) in methylene chloride (350 ml)-methanol (150 ml) was added at 0° C. and the mixture was then stirred at room temperature for 20 minutes. To this reaction mixture was added an aqueous solution of sodium sulfite and the mixture was extracted with methylene chloride. The extract was washed with saturated brine, dried over dry sodium sulfate, and filtered. The filtrate was concentrated under reduced pressure to provide 2-benzyloxymethyl-5-iodo-4-isopropylimidazole (4c). Yield 93 g (85.5%).
