Procedure
Sephadex G-25 eluates were prepared in 0.1 M Tris-HCl, pH 7.8, 5 mM EDTA, 0.3 M KCl, and 10% (v/v) glycerol. The eluate was incubated with 2 mM GTP for 90 min at 37° C. in the dark. The dihydroneopterin triphosphate formed was oxidized to neopterin triphosphate with 0.1 M HCl, 0.01 M I2 for 1 hour in the dark at room temperature. Precipitate was removed by centrifugation at 10,000×g for 5 min. Excess iodine was reduced by 0.1 M ascorbic acid and the acid neutralized by adding 1 M NaOH. Neopterin was produced by incubation with 8 Units/ml of alkaline phosphatase for 1 hour at 37° C. Neopterin was quantified by reverse-phase HPLC with fluorescence detection.
