To a solution of tert-butyl 3-(6-amino-7-(4-chlorophenyl)-8-oxo-7H-purin-9(8H)-yl)phenylcarbamate (12) (234 mg, 0.52 mmol) in DCM (8 mL) was added TFA (2 mL) dropwise. The reaction mixture was stirred at rt for 1 h before concentrated. The residue was purified by column chromatography (silica gel, 0 to 5% methanol in methylene chloride (0.3% Et3N)) to give 6-amino-9-(3-aminophenyl)-7-(4-chlorophenyl)-7H-purin-8(9H)-one as a brown solid (13) (180 mg, 98.9%). LC-MS (ESI): m/z (M/M+2) 353/355.
