The cyanocarbamimidate from step A (0.47 g, 1.7 mmol), (S)-5-methyl-4-(3-methylpiperazin-1-yl)-7H-pyrrolo[2,3-d]pyrimidine from Example 1, step B (0.40 g, 1.7 mmol), and N,N-diisopropylethylamine (1 ml) were added to acetonitrile (10 ml). The mixture was heated at 85° C. in a sealed pressure tube for 4 hours. The solvent was evaporated, and the residue was purified by flash chromatography (80 g SiO2, 0-5% MeOH:CH2Cl2, 50 min) to give (S)—N-(3-chlorophenyl)-N′-cyano-2-methyl-4-(5-methyl-7H-pyrrolo[2,3-d]pyrimidin-4-yl)piperazine-1-carboximidamide (0.32 g, 0.79 mmol, 46%).
