Procedure

To 478.7 mg (1.60 mmol) of 2-[(thieno[2, 3-c]pyridin-7-yl)methylthio]benzimidazole. 1/10 H2O (Ia-1) were added 8 ml of CH3CN and 324.6 mg (4.00 mmol) of 37% aqueous solution of HCHO, and the mixture was stirred for 15 minutes at 70° C. After evaporation the solvent under reduced pressure, the residual crystals were washed with ether, whereby 420.9 mg (Yield : 80.3%) of the objective compound (Ib-4), 1-hydroxymethyl-2-[(thieno[2, 3-c]pyridin-7-yl)methylthio]benzimidazole was obtained as crystals.

Workup

  1. customAfter evaporation the solvent under reduced pressure
  2. washthe residual crystals were washed with ether, whereby 420.9 mg (Yield : 80.3%) of the objective compound (Ib-4)
