Activity on cellobiose and 4-nitrophenyl-beta-D-glucopyranoside: Cellobiose was dissolved in 50 mM sodium acetate-0.01% TWEEN® 20 pH 5 buffer to 2 g/L. Enzyme was diluted at various concentrations using the same buffer. To 50 μl of the enzyme dilutions (0.04-5 μg/ml), 100 μl of the cellobiose solution (2 g/L buffer) was added. The reaction was incubated at 50° C. for 30 minutes before the reaction was stopped with 50 μl of 0.5 M sodium hydroxide. Glucose produced was measured using the Glucose Oxidase reagent (Pointe Scientific, Inc., Canton, Mich., USA) according to the manufacturer.
