A tetrahydrofuran solution (6 ml) of (5-bromo-2-methylsulfanyl-pyrimidin-4-yl)-(S)-piperidin-3-yl-amine (from Example 76, step 1 above) (300 mg, 0.99 mmol), diazabicycloundecane (530 mg, 3.5 mmol) and ethyl 2-cyanoacetate was stirred at room temperature for 20 hours. The reaction was diluted with water and extracted with ethyl acetate (2×). The organics were dried and evaporated to give 3-[(S)-3-(5-bromo-2-methylsulfanyl-pyrimidin-4-ylamino)-piperidin-1-yl]-3-oxo-propionitrile as an oil. MS (ES+): 371.
