3-(2-(Tetrahydro-2H-pyran-4-yl)acetyl)dihydro-2H-pyran-4(3H)-one (200 mg, 0.88 mmol), 1-(4-(2-methyl-1H-imidazol-1-yl)phenyl)guanidine (190 mg, 0.88 mmol, Example 41c) and potassium carbonate (122 mg, 0.88 mmol) was slurrified in EtOH (4 mL) and heated to 50° C. for 15 h. DCM and water were added. The organic phase was separated, dried with MgSO4 and the solvent was evaporated. The crude product was purified using preparative HPLC yielding N-(4-(2-methyl-1H-imidazol-1-yl)phenyl)-4-((tetrahydro-2H-pyran-4-yl)methyl)-7,8-dihydro-5H-pyrano[4,3-d]pyrimidin-2-amine (15.0 mg, 4.0%).
