(R)-tert-Butyl 2-(4-(2-methyl-1H-imidazol-1-yl)phenylamino)-4-((tetrahydrofuran-2-yl)methylamino)-7,8-dihydropyrido[4,3-d]pyrimidine-6(5H)-carboxylate (75 mg, 0.15 mmol) was dissolved in methanol (2 mL). Hydrochloric acid (4.51 μL, 0.15 mmol) was added and the reaction mixture was stirred at 75° C. for 30 minutes. The solvent was evaporated under reduced pressure and the crude (R)-N2-(4-(2-methyl-1H-imidazol-1-yl)phenyl)-N4-((tetrahydrofuran-2-yl)methyl)-5,6,7,8-tetrahydropyrido[4,3-d]pyrimidine-2,4-diamine (56.0 mg, 93%) was used as such in the subsequent step.
