Procedure
HCl (Sigma Aldrich Chemical Company, Inc., Milwaukee, Wis., USA 2M in ether; 0.43 mL, 0.86 mmol) was added to a solution of 4-[4-(2-Methyl-pyridin-3-yloxy)-pyrazolo[3,4-d]pyrimidin-1-yl]-piperidine-1-carboxylic acid isopropyl ester (170 mg, 0.43 mmol) in anhydrous ether (2 mL). The resulting mixture was shaken at room temperature for 10 min. The solvent was evaporated under high vacuum to give a white powder which still contained some ether by NMR. The solid was ground to a fine powder using a spatula and then it was held overnight in a vacuum oven overnight at 80° C. to give 4-[4-(2-methyl-pyridin-3-yloxy)-pyrazolo[3,4-d]pyrimidin-1-yl]-piperidine-1-carboxylic acid isopropyl ester hydrochloride salt (147 mg, 79%) as a white powder. 1H NMR (300 MHz, CDCl3) δ 1.28 (d, 6H, J=6.0 Hz), 2.01-2.04 (m, 2H), 2.22-2.31 (m, 2H), 2.87 (s, 3H), 2.98-3.06 (m, 2H), 4.30-4.42 (m, 2H), 4.95-5.05 (m, 2H), 7.79-7.84 (m, 1H), 8.19-8.24 (m, 2H), 8.44 (s, 1H), 8.61-8.63 (m, 1H).
