A mixture of 5A (80 mg, 0.245 mmol), 1-isopropyl-1H-pyrazol-3-amine (36.8 mg, 0.294 mmol), Pd2(dba)3 (11.23 mg, 0.012 mmol), 9,9-Dimethyl-4,5-bis(diphenylphosphino)xanthene (7.10 mg, 0.012 mmol) and Cs2CO3 (160 mg, 0.491 mmol) in dioxane (2.0 mL) was degassed with argon. The mixture was heated at 110° C. under a nitrogen atmosphere overnight. After cooling to room temperature, the mixture was concentrated to remove dioxane and then water was added. The resulting precipitate was collected by filtration, rinsed with water and dried to give crude 48A (94 mg, 92%) as a dark brown solid. HPLC Rt=3.416 min (Chromolith SpeedROD 4.6×50 mm, 10-90% aqueous methanol containing 0.1% TFA, 4 min gradient, monitored at 220 nm), [M+H]=415.
