Reaction Steps

10-formyltetrahydrofolate-synthetase catalyzes the reaction, formate+THF+ATP→10-formyl-THF+ADP+Pi. A test tube containing 100 μL 1 M triethanolamine buffer (pH 7.5), 100 μL 1 M ammonium formate (pH 7.5), 100 μL 25 mM MgCl2, 100 μL 2 M KCl, 100 μL 10 mM THF, freshly made 100 μL 20 mM ATP, and 390 μL water was pre-incubated for 2 min at 30° C. A 10 μL aliquot of the extracted sample containing enzyme was added and incubated 15 min at 30° C. The reaction was terminated by adding 2 mL 0.36 N HCl. The 10-formyl-THF formed during the incubation period was converted to 5,10-methenyl-THF by the acid. The absorbency was measured at 350 nm against a water blank and the 5,10-methenyl-THF formed was determined by using an extinction coefficient of 24,900 M-1 cm-1.
