Procedure

The tert-butyl 3-amino-5-hydroxy-4,6-dimethyl-thieno[2,3-b]pyridine-2-carboxylate obtained above (12 g, 40 mmol) was dissolved in CH3CN (300 mL), followed by the addition of K2CO3 (11 g, 80 mmol) and 1-bromo-2-chloroethane (5.7 g, 40 mmol). The mixture was heated to reflux for 8 h, water (100 mL) was added and the mixture was extracted with ethyl acetate (60 mLx 3). The organic layers were washed with 0.5% aqueous sodium hydroxide (150 mL) and brine, dried over anhydrous Na2SO4 and then concentrated to give tert-butyl 3-amino-5-(2-chloroethoxy)-4,6-dimethyl-thieno[2,3-b]pyridine-2-carboxylate as a yellow solid (6.9 g, 50%).
