Treat a 22° C. solution of 4-{5-[3-(5-tert-butyl-2-p-tolyl-2H-pyrazol-3-yl)-ureido]-pyridin-2-ylamino}-piperidine-1-carboxylic acid tert-butyl ester (1.1 mmol, 0.6 g) in Et2O (5 mL) with HCl (2.0 M solution in diethyl ether, 5 mL, 10 mmol). Stir the solution overnight at room temperature. Concentrate under reduced pressure to give a residue, then subject residue to SCX cartridge eluting with ammonia 2.0 N in methanol. Obtain the title compound as the free base. MS(ES): m/z=488 [M+H].
