A 2.0 M HCl solution in Et2O (212 μL, 0.42 mmol) was slowly added at room temperature to a stirring solution of 3-hydroxymethyl-4-[2-(2-hydroxy-phenyl)-7-methyl-quinazolin-4-yl]-piperazine-1-carboxylic acid benzyl ester (206 mg, 0.42 mmol) in 500 μL of CH2Cl2. The reaction was stirred for 1 hour. Solvents were removed under reduced pressure, and the residue was triturated with Et2O and filtered to give benzyl 3-(hydroxymethyl)-4-(2-(2-hydroxyphenyl)-7-methylquinazolin-4-yl)piperazine-1-carboxylate hydrochloride. LC/MS: m/z 485.5 (M+H)+ at 3.07 min (10%-99% CH3CN (0.035% TFA)/H2O (0.05% TFA))
