Procedure
To a solution of compound 451 (170 mg, 0.51 mmol) in ethanol (2 mL) was added but-3-yn-1-ol (39 μL, 0.51 mmol). A freshly prepared solution of copper(II) sulphate pentahydrate (5.0 mg, 0.020 mmol) and sodium ascorbate (20 mg, 0.10 mmol) in water (0.5 mL) was added to the reaction mixture. The flask was closed and stirred for 24 h at RT under argon. The reaction mixture was poured into a mixture of EtOAc/water. The organic phase was washed with water, brine and then dried (MgSO4), filtered and concentrated in vacuo to give the crude product. The crude product was purified by continuous gradient flash chromatography using EtOAc/petroleum ether (40-60) 40:60 to 95:5 as the eluent to afford the title compound as white syrup.
