Procedure
To a solution of compound 455 (1.32 g, 3.56 mmol) in ethanol (20 mL) was added but-3-yn-1-ol (0.3 mL, 3.91 mmol). A freshly prepared solution of copper(II) sulphate pentahydrate (36 mg, 0.14 mmol) and sodium ascorbate (141 mg, 0.71 mmol) in water (3.2 mL) was added to the reaction mixture. The flask was closed and stirred for 24 h at RT under argon. The reaction mixture was poured into a mixture of EtOAc/water. The organic phase was washed with water, brine and then dried (MgSO4), filtered and concentrated in vacuo to give the crude product. The crude product was purified by continuous gradient flash chromatography using EtOAc/petroleum ether (40-60) 40:60 to 95:5 as the eluent to afford the title compound as white syrup.
