To a slurry of 2-amino-nicotinamide (100 mg, 0.7 mmol) in THF (5 ml) was added dropwise benzyloxyacetyl chloride [19810-31-2] (127 μL, 0.8 mmol) in THF (2 ml) at 5° C. The reaction mixture was heated to reflux for 2 hours. Water was added, the phases were separated and the inorganic layer was extracted with EtOAc (3 times). The combined organic layers were washed with brine, dried over Na2SO4, filtrated and concentrated in vacuo. Column chromatography on silica gel with CH2Cl2:MeOH 9:1 as eluent yielded 2-(2-benzyloxy-acetylamino)-nicotinamide (76 mg, 36%), MS(m/e): 286.1 [M+H+].
