Methyl 5-(N-(5-cyclopropyl-3-(methylcarbamoyl)-2-p-tolylfuro[2,3-b]pyridin-6-yl)methylsulfonamido)pentanoate (17 mg, 0.033 mmol) was dissolved in MeOH/THF/H2O (2.33 mL, 3:1:3) and LiOH (0.13 mL, 0.13 mmol, 1M solution) was added and the mixture stirred at 23° C. overnight. The reaction was then neutralized by addition of NH4Cl and the aqueous extracted with EtOAc (3×10 mL) then washed with brine and dried over Na2SO4 and evaporated to dryness. HPLC (Basic method/NH4OH/CH3CN/H2O) yielded 5-(N-(5-cyclopropyl-3-(methylcarbamoyl)-2-p-tolylfuro[2,3-b]pyridin-6-yl)methylsulfonamido)pentanoic acid (5 mg, 0.01 mmol, 30%).
