Procedure
(2,4-Diamino-5-fluoro-phenyl)-acetic acid 3c (7.12 g, 38.7 mmol) was dissolved in hydrochloric acid (100 ml, 1 mol/L) under stirring at room temperature. The solution was heated to reflux for 1 hour. After thin lay chromatography showed the disappearance of starting materials, the reaction mixture was cooled down to room temperature, neutralized with sodium hydroxide solution (100 ml, 1 mol/L) in an ice-water bath, extracted with ethyl acetate (125 ml×4). The combined organic extracts were washed with saturated brine (100 ml), dried over anhydrous magnesium sulfate, filtered to remove the drying agent and concentrated under reduced pressure to obtain the title compound 5-fluoro-6-amino-indol-2-one 3d (5.3 g, yield 82.8%) as a yellow solid.
