Tert-butyl4-[(3,6-dichloro-2-{[(4,6-dimethyl-2-oxo-1,2-dihydropyridin-3-yl)methyl]carbamoyl}pyridin-4-yl)(methyl)amino]piperidine-1-carboxylate (310 mg, 1.29 mmol) was dissolved in 4M HCl in dioxane solution (3 ml) and stirred at room temperature for 16 h after which the solvent was evaporated and the compound purified by preparative HPLC to afford the title compound as a beige powder (13 mg, 14%). LC-MS 100%, 2.41 min (7 minute LC-MS method), m/z=438.1/439.8, 1H NMR (500 MHz, Methanol-d4) δ 7.12 (s, 1H), 6.11 (s, 1H), 4.46 (s, 2H), 4.01-3.81 (m, 1H), 3.45 (d, J=12.9 Hz, 2H), 3.12-2.97 (m, 2H), 2.82 (s, 3H), 2.35 (s, 3H), 2.22 (s, 3H), 2.13-1.93 (m, 4H).
