Procedure

A solution of ethyl 5-(4-{[(3,4-dichloro-5-methyl-1H-pyrrol-2-yl)carbonyl]amino}piperidin-1-yl)-1,3,4-thiadiazole-2-carboxylate (Example 219, 280 mg, 0.65 mmol) in THF (5 ml) was cooled to 0° C. and treated with diisobutylaluminium hydride (1.3 ml, 1.95 mmol) dropwise over 5 min. The reaction was kept in the ice bath and allowed to slowly warm to room temperature while stirring overnight. LC-MS indicated that the reaction was complete. The reaction was quenched with aqueous Rochelle salt, and the resultant mixture was diluted with EtOAc (10 ml) and stirred vigorously for 2 h. The layers were separated, and the aqueous portion was extracted with EtOAc. The combined organic portions were dried (Na2SO4), filtered, and concentrated to give a pale orange oil. The crude material was purified by preparatory HPLC (35-75% CH3CN/H2O, 0.1% TFA, 14 min.) to give 17 mg of the title compound.
