To a mixture of 5-bromo-2-(3-ethylureido)pyridine-4-carbothioamide (Intermediate 42, 1 g, 3.30 mmol) in ethanol (25 mL), 1-bromobutan-2-one (0.337 mL, 3.30 mmol) was added, and the reaction mixture was heated to 80° for 4 h, then cooled to room temperature, concentrated under reduced pressure, and the resulting solid was washed with acetonitrile to give the title compound as an off-white solid (431 mg). MS (ESP): 356 (M+1) for C13H15BrN4OS; NMR: 1.09 (t, 3H); 1.29 (t, 3H); 2.84 (q, 2H); 3.15-3.20 (m, 2H); 7.34 (br s, 1H); 7.66 (s, 1H); 8.38 (s, 1H); 8.50 (s, 1H); 9.36 (s, 1H).
