To test the bound aminoacylase activity, 100 mg of the charged carrier were carefully shaken for 2 minutes at room temperature in 3 ml of 0.1 molar phosphate buffer solution (pH 7.6) containing 10 mg/ml of N-acetyl-D,L-methionine and 0.36 mg of cobalt chloride (CoCl2 ·6 H2O). The amount of L-methionine formed was then found by reaction with L-aminoacid-oxidase and determination of the resulting hydrogen peroxide with peroxidase by UV-spectrophotometric measurement of o-dianisidine (in the oxidized, ie. quinoid, form) (Methods in Enzymology, Vol. II).
