Procedure

To a stirred solution of 332 (103 mg, 0.227 mmol) in DCM (2.2 mL) at RT under nitrogen was added TFA (1 mL). After 2 h the reaction was concentrated to dryness in a rotovap and high vacuum. The crude presumed TFA salt was redissolved in DCM (5 mL) and stirred rapidly with 10% aqueous Na2CO3 (5 mL). After 5 min the organics phase was separated, dried (MgSO4), filtered and concentrated to afford 80 mg (100%) of (1S,2R)-1-amino-1-(3-chloro-4-fluorophenyl)propan-2-yl 4-nitrobenzoate (334) as a yellow oil which was used without additional purification.
