Procedure
To a 2-mL microwave vial was added 2-fluoro-6-(2-furanyl)-4-{2-(methylamino)-6-[(3R)-3-methyl-4-morpholinyl]-4-pyrimidinyl}benzonitrile (50 mg, 0.127 mmol), hydrazine (0.044 mL, 0.763 mmol), Hunig's base (0.067 mL, 0.381 mmol) and ethanol (1 mL). The reaction was heated at 150° C. for 90 minutes in a microwave reactor. The reaction mixture was allowed to cool, then was concentrated in vacuo, and the residue was purified by flash chromatography (0-10% CH3OH in CH2Cl2) to afford the title compound (35 mg, 0.086 mmol) as a yellow solid. LC-MS (ES) m/z=406 [M+H]+. 1H NMR (400 MHz, DMSO-d6): δ 1.20 (d, J=6.6 Hz, 3H), 2.75-2.94 (m, 3H), 3.05-3.21 (m, 1H), 3.41-3.52 (m, 1H), 3.57-3.67 (m, 1H), 3.68-3.80 (m, 1H), 3.87-4.00 (m, 1H), 4.06-4.21 (m, 1H), 4.45-4.62 (m, 1H), 5.11 (s, 2H), 6.46-6.68 (m, 2H), 6.68-6.74 (m, 1H), 6.97-7.16 (m, 1H), 7.74-7.82 (m, 1H), 7.89-7.95 (m, 1H), 7.97-8.07 (m, 1H), 11.95 (s, 1H).
