Procedure
5 mg of 8-propylcarboxytheophylline dissolved in 100 μl of dimethylformamide (DMF) was mixed with 3 mg of N-hydroxysuccinimide and 5 mg of water-soluble carbodiimide, and stirred at room temperature for 2 hours to obtain activated theophylline. 500 μl of the above activated theophylline solution was added to 10 mg of horse ferritin dissolved in 1 ml of 0.1M sodium hydrogen carbonate aqueous solution, and allowed to stand at room themperature for 1 hour. Unreacted substances were removed by using a Sephadex G-25 gel column previously equilibrated with a phosphate-buffered saline solution of pH 7.0 to obtain 9 mg of the object macromolecular antigen of horse ferritin-theophylline conjugate.
