Under an N2 atmosphere, a 1.0 M solution of HCl in ether (0.53 mL, 0.53 mmol) was added dropwise to a solution of (R)-ethyl 1-(2-(2-hydroxyphenyl)-7-methylquinazolin-4-yl)pyrrolidin-3-ylcarbamate (208 mg, 0.53 mmol) in CH2Cl2 (13 mL). After stirring the reaction for 10 minutes, ether (30 mL) was added, and a precipitate formed which was filtered and dried to give (R)-ethyl 1-(2-(2-hydroxyphenyl)-7-methylquinazolin-4-yl)pyrrolidin-3-ylcarbamate hydrochloride (HCl salt of compound 2). 1H NMR (400 MHz, DMSO-d6) δ 8.29-8.23 (m, 2H), 7.78 (s, 1H), 7.61 (d, J=5.1 Hz, 1H), 7.52-7.48 (m, 2H), 7.10 (d, J=8.1 Hz, 1H), 7.06-7.02 (m, 2H), 4.29-4.13 (m, 4H), 4.03-3.94 (m, 3H), 2.54 (s, 3H), 2.27-2.22 (m, 1H), 2.08-2.06 (m, 1H), 1.16 (t, J=7.0 Hz, 3H) ppm. LC/MS: m/z 393.3 (M+H)+ at 2.36 min (10%-99% CH3CN (0.035% TFA)/H2O (0.05% TFA)).
