To a solution of [(S)-1-(4-methyl-1-phenyl-1H-benzoimidazol-2-yl)ethyl]carbamic acid tertbutyl ester (380 mg, 1.08 mmol) in DCM (5 mL) was added TFA (1 mL) and the mixture was stirred at RT for 3 h. Additional TFA (0.5 mL) was added and stirring was continued for 30 min. Volatiles were then removed under reduced pressure and the residue was then dissolved in a small amount of DCM and loaded onto an SCX-2 cartridge which was initially washed with 10% MeOH in DCM. The product was eluted with 2M NH3/MeOH affording (S)-1-(4-Methyl-1-phenyl-1H-benzoimidazol-2-yl)ethylamine as a brown solid (222 mg, 82%). LCMS: RT 2.16 [M-NH2]+ 235.1
