Procedure
Glucose and ethanol were determined by high-performance liquid chromatography (HPLC). Fermentation broth was filtered through a 0.45 μm nylon filter, diluted 10-fold with 0.01N H2SO4, and injected into a Bio-Rad Aminex HPX-87H organic acid column (Waters Chromatography, Milford, Minn.) maintained at 60° C. The components were eluted from the column with 0.01N H2SO4 at a flow rate of 0.5 ml/min. The separated compounds were detected and quantified with a Beckman refractive index detector (Model 156). Known concentrations of compounds were used as standards.
