A solution of 3-aminothiophene-2-carboxamide (0.90 g, 6.3 mmol), Et3N (0.97 mL, 7.0 mmol), and dimethylaminopyridine (0.15 g, 1.3 mmol) in CH2Cl2 (21 mL) was cooled in an ice bath, and 2-methoxybenzoyl chloride (1.1 g, 6.3 mmol) was added dropwise. The reaction was stirred for 1 h, and water (20 mL) was added. The solids were filtered, washed with water, and dried under vacuum to give 3-(2-methoxybenzamido)thiophene-2-carboxamide (1.4 g, 79% yield). LC/MS: m/z 277.3 (M+H)+ at 2.61 min (10%-99% CH3CN (0.035% TFA)/H2O (0.05% TFA)).
