2-{2-[4-(4-chloro-3-trifluoromethyl-phenoxy)-phenyl]-ethylamino}-1H-pyrimidin-4-one (0.122 mmol, 1 eq), paraformaldehyde (3.66 mg, 0.112 mmol, 1 eq) and pyrrolidine (10 μl, 0.112 mmol, 1 eq) were refluxed for 90 mins. Solvent was evaporated in vacuo to give 2-{2-[4-(4-chloro-3-trifluoromethyl-phenoxy)-phenyl]-ethylamino}-5-pyrrolidin-1-ylmethyl-1H-pyrimidin-4-one (0.089 mmol, yield: 73%, UPLC-MS/UV:[M+H]+=493.39; rt: 0.98 min; purity: 85%). 1H NMR (300 MHz; CDCl3) δ/ppm 1.88-1.91 (m, 4H), 2.68-2.76 (m, 4H), 2.90 (t, J=7.51, 2H), 3.60 (s, 2H), 3.61-3.68 (m, 2H), 6.89-6.96 (m, 2H), 7.02 (dd, J=8.93 J=3.12, 1H), 7.21-7.23 (m, 1H), 7.30 (d, J=2.97, 1H), 7.40 (d, J=9.07, 1H), 7.74 (s, 1H)
