Procedure

A mixture of 2-(3-chloro-4-fluorobenzyl)-4-(chloromethyl)-10-methoxy-8-methyl-7,8-dihydropyrazino[1′,2′:1,5]pyrrolo[2,3-d]pyridazine-1,9(2H,6H)-dione (0.16 g, 0.37 mmol; Example 100) and sodium cyanide (30 mg, 0.61 mmol) in anhydrous DMF (2 mL) was heated in a sealed tube at room temperature overnight. The product mixture was concentrated under vacuum. The residue was partitioned between dichloromethane and brine. The organic extract was washed with brine, dried over anhydrous magnesium sulfate, filtered, and concentrated under vacuum. The residue was subjected to column chromatography on silica gel eluting with a 0 to 10% methanol in chloroform gradient. Collection and concentration of appropriate fractions afforded the title compound.
