Procedure
A mixture of tert-butyl (1S,2R)-2-(6-(2,4-dimethoxybenzyl)-4-(1,5-dimethyl-1H-pyrazol-4-yl)-5-oxo-6,7-dihydro-5H-pyrrolo[3,4-d]pyrimidin-2-ylamino)cyclohexylcarbamate obtained above in TFA (2 mL) was stirred at 70° C. for 2 h. The mixture was subsequently purified by reverse phase preparative HPLC. The fractions were collected and concentrated to give a TFA salt of the title compound as a white solid (0.5 mg). 1H NMR (500 MHz, CD3OD) δ ppm 1.59 (br s, 2 H), 1.67-2.02 (m, 6 H), 2.66-2.72 (m, 3 H), 3.67 (br s, 1 H), 3.85 (s, 3 H), 4.29 (s, 2 H), 4.50-4.64 (m, 1 H), 8.64 (br s, 1 H). [M+H] calc'd for C17H23N7O, 342; found, 342.
