Following general procedure IV-2, (R)-tert-butyl [1-(5-{[3-(cyclopropanecarbonyl)-6-(3-chloro-5-fluoro-4-hydroxyphenyl)-1,5-naphthyridin-4-yl)amino)pyridin-2-yl)piperidin-3-yl)carbamate (0.98 g, 0.16 mmol) was reacted with TFA (2 mL). The resulting trifluoroacetate salt of the product was converted to the free base to afford the desired product (58 mg, 71%) as an orange solid: 1H NMR (500 MHz, CD3OD) δ 9.18 (s, 1H), 8.09 (d, J=9.0 Hz, 1H), 8.04 (d, J=9.0 Hz, 1H), 7.97 (d, J=2.0 Hz, 1H), 7.37 (dd, J=9.0, 2.0 Hz, 1H), 6.97 (d, J=13.0 Hz, 1H), 6.75 (d, J=9.0 Hz, 1H), 4.18-4.15 (m, 1H), 3.83-3.81 (m, 1H), 3.31-3.22 (m, 1H), 3.15-3.05 (m, 2H), 2.91-2.85 (m, 1H), 2.12-2.08 (m, 1H), 1.91-1.83 (m, 1H), 1.71-1.58 (m, 2H), 1.25-1.08 (m, 4H); ESI MS m/z 533 [M+H]+; HPLC 99.0% (AUC), tR=9.18 min.
