Procedure
6-methyl-2-({[1-(propane-2-yl)-1H-benzimidazol-2-yl]methyl}sulfanyl)pyrimidin-4-ol (500 mg, 1.6 mmol) was stirred in methanol (50 mL) and a solution of 4 N HCl in dioxane (600 μL, 2.4 mmol) was added dropwise at 0° C. The mixture was stirred for 1 hour at room temperature. The solvent was removed by evaporation, and the residue was triturated with diethyl ether and dried in vacuo to afford 6-methyl-2-({[1-(propane-2-yl)-1H-benzimidazol-2-yl]methyl}sulfanyl)pyrimidin-4-ol hydrochloride (560 mg, 100% yield); 1H NMR (400 MHz, DMSO-d6): δ 1.68 (s, 3H), 1.70 (s, 3H), 2.14 (s, 3H), 5.01 (s, 2H), 5.21-5.28 (m, 1H), 6.20 (s, 1H), 7.54-7.61 (m, 2H), 7.84-7.87 (m, 1H), 8.16-8.19 (m, 1H); LRMS (ES+) m/z 315 (100%, M+1).
