n-BuLi (1.65 mL, 4 mmol) was added dropwise to a mixture of 4-bromo-1-trityl-1H-pyrazole (1.5 g, 3.8 mmol) in THF (15 mL) cooled to −78° C. Then the mixture was stirred at −78° C. for 1 hour. After methyl 4-acetylbenzoate (0.71 g, 3.8 mmol) in THF (1 mL) was added, the mixture was stirred at −78° C. for 2 hours, and then quenched with saturated aqueous ammonium chloride solution. The mixture was extracted with ethyl acetate. The organic layer was concentrated and the residue was purified by chromatography on silica gel to afford methyl 4-[1-hydroxy-1-(1-trityl-1H-pyrazol-4-yl)ethyl]benzoate. MS ESI calc'd. for C32H29N2O3 [M+H]+ 489. found 489.
