Procedure
To a mixture of 1-cyclopentyl-3-ethyl-6-(4-aminophenyl)pyrazolo[3,4-d]pyrimidin-4-one hydrochloride 1/3 hydrate (0.61 g, 1.7 mmol) in dry pyridine (20 ml) in an ice bath was added methanesulfonyl chloride (0.49 g, 4.25 mmol). The reaction mixture was stirred at room temperature for about two days, the solvent was stripped to dryness and the residue was treated with water. The product was collected by filtration and recrystallized from ethanol, after DARCO® treatment, to afford 0.49 g (72%) of 1-cyclopentyl-3-ethyl-6-[4-(methylsulfonylamino)phenyl]pyrazolo[3,4-d]pyrimidin-4-one, m.p. 325° C. (dec.).
