To a suspension of 6-aminouracil (15 g, 0.118 mol) in H2O (600 mL) was added sodium acetate (NaOAc) (15 g, 0.183 mol) and chloroethanal (ClCH2CHO) (50% in H2O, 30 mL). The mixture was heated at reflux for 4 h and then cooled to room temperature. The resulting dark brown precipitates were collected by filtration to afford crude 7H-Pyrrolo[2,3-d]pyrimidine-2,4-diol (14.4 g, 81% yield).
