A mixture of 4-methoxy-5,6,7,8-tetrahydropyrido[4,3-d]pyrimidine (12.88 g, 0.078 mol), 5-chloro-2-fluorobenzonitrile (40 g, 0.2 mol), and 1,8-diazabicyclo[5.4.0]undec-7-ene (60 mL, 0.4 mol) was heated at 80° C. for 3 hours. After cooling the mixture was diluted with CH2Cl2 (300 mL), washed with water, dried, and concentrated to afford a brown solid which was purified by silica gel chromatography (0-50% EtOAc/hexane) to afford a solid (19.33 g, 78.3%).
